Adherence patterns of diarrheagenic Escherichia coli strains isolated from children with gastrointestinal diarrhea and matched controls in a Hep-2 cell adhesion assay

چکیده
Escherichia coli is considered as the main cause of epidemic outbreaks and endemic cases of child diarrhea. In the present study, the capacity of a large collection of Escherichia coli (E. coli) strains and matched controls was investigated to determine their adherence pattern on Hep-2 cell lines. A total of 66 pure E. coli strains, which were previously isolated from children aged less than 12 years with diarrhea, were selected for adherence assay. Moreover, 2 E. coli strains, with known adherence patterns, obtained from the Persian Type Culture Collection were used as the control. Hep-2 cell monolayers (50 to 70% confluence) were grown on glass cover slips. Subsequently, 20 μl of bacterial cultures (2 × 106 cfu/ml bacteria) was added into each well, and the plates were incubated in a humid atmosphere with 5% CO2 for 3 hours. Then, culture medium was aspirated from the monolayers, which were washed 4 times with phosphate-buffered saline, fixed in 70% aqueous methanol for 5 minutes, stained with Giemsa stain, and examined using light microscopy. In total, 66 E. coli strains were evaluated for their adherence pattern on Hep-2 cell line. All the isolates adhered to the Hep-2 cells with the exception of 3. The other 63 showed aggregative adherence (AA) (34%), localized adherence (LA) (31%), or diffuse adherence (DA) (29%) patterns. The present investigation revealed that this assay is convenient and economical for identification of a large number of diarrheagenic E. coli isolates’ adherence to Hep-2 cell lines.

In the present study, carbon species including organic carbon (OC), elemental carbon (EC), and water-soluble organic carbon (WSOC) concentration in PM2.5 were assessed at an urban site of Tehran, Iran during March to June 2014. The PM2.5 samples were collected using an frmOMNITM Ambient Air Sampler. Thermal gravimetric analysis (TGA) was used to analyze OC and EC. The results showed that PM2.5 ...
متن کاملEvaluation of a simplified HEp-2 cell adherence assay for Escherichia coli isolated from south Indian children with acute diarrhea and controls.
A simplified HEp-2 cell adhesion assay was performed with stored Escherichia coli isolates from 761 children with acute diarrhea and 531 matched controls, and the results were evaluated by means of fluorescent actin staining and hybridization with DNA probes. The prevalence rates of localized adherence and aggregative adherence were significantly higher for patients (9.9 and 7.6%, respectively)...
متن کاملA localized adherence-like pattern as a second pattern of adherence of classic enteropathogenic Escherichia coli to HEp-2 cells that is associated with infantile diarrhea.
Escherichia coli strains that cause nonbloody diarrhea in infants are known to present three distinct patterns of adherence to epithelial cells, namely, localized (LA), diffuse (DA), and aggregative (AA) adherence. Strains with LA (typical Enteropathogenic Escherichia coli [EPEC]) are well recognized as a cause of secretory diarrhea, but the role of strains with DA (DAEC) is controversial, and ...

The adherence property of 70 verotoxigenicE. coli (VTEC) strains, not belonging to O157: H7 serotype (non-O157) was checked on HeLa cells adherence assay. These strains were isolated from patients with diarrhea and healthy individuals. The results obtained in this study revealed that localised adherence (LA) was manifested by 3 strains of which two belonged to serogroups O111, O127 and the thir...
متن کاملHep-2–Adherent Escherichia coli Strains Associated with Acute Infantile Diarrhea, São Paulo, Brazil
In this paired case-control study of infants with diarrhea in São Paulo, we examined the association between HEp-2-adherent Escherichia coli strains and diarrhea. We tested isolates from stool specimens of infants with diarrhea and matched controls in an HEp-2 cell adherence assay; we then hybridized isolates with DNA probes and identified enteropathogenic E. coli (EPEC), enteroaggregative E. c...
